TempO-Seq
Gene expression analysis platforms have evolved from outdated hybridisation array based technologies and RT-qPCR to highly multiplexed RNA-Seq de novo discovery platforms. Although RNA-Seq is commonly used as a quantitative assay to determine relative transcript abundance, it is costly, onerous, and employs a time intensive process for assay design, running the assay and data analysis. To address these challenges, BioSpyder Technologies has developed a novel product for targeted sequencing called TempO-Seq®. TempO-Seq is a high-throughput assay specifically designed for very efficient profiling of molecular signatures or transcriptomes. Importantly, based on its underlying molecular biology lacking the need for nucleic acid purification or reverse transcription, TempO-Seq excels where traditional techniques fail: low input, RNA degradation, or formalin fixation have negligible impact on assay performance. Therefore, TempO-Seq is ideally suited for molecular profiling and gene expression from FFPE sections, tissue microarrays, or fine needle aspirates. Similarly, based on its scalability, TempO-Seq can profile transcriptomes of thousands of samples at once, making an ideal (additional) readout for toxicogenomics or drug discovery.

Sample input
TempO-Seq allows for high flexibility in terms of sample input type, quantity, and quality. The TempO-Seq assay requires only very low amounts of input material and therefore allows for the molecular analysis of precious samples. The assay is furthermore insensitive to RNA degradation (down to RIN 1-3) or cross-linking and fragmentation resulting from formaldehyde fixation [1]. This enables us to run the assay directly from small, pre-defined areas on FFPE slides.
We routinely process thousands of samples a week, either from cell lysates or FFPE tissue.
We have also established protocols to assay tissue microarrays, single cell lysates, or liquid biopsies using TempO-Seq.

Assay specifics & multiplexing
TempO-Seq is a highly efficient multiplexed molecular profiling platform. TempO-Seq targeted RNA expression or DNA variant assays are designed to monitor tens to tens of thousands of genes or markers (single nucleotide variants and indels). We are also able to assess proteins, and can measure RNA, DNA, and protein in the same sample & reaction. TempO-Seq benefits from extremely high analytical sensitivity (down to single-cell level) and specificity (99.8% for a single base mismatch), enabling precise studies from very limited sample input.
TempO-Seq allows for quick and inexpensive high-throughput profiling of the whole-protein coding transcriptome of ~20,000 genes which enables high-throughput studies with transcriptomic endpoints, for example in drug discovery projects.
Due to its low price and ability to process tissue from a wide range of sources, TempO-Seq is also ideal for enabling the day-to-day implementation of transcriptomics for personalised diagnostics in the clinic. Several diagnostic tests are currently in our development pipeline, and we are always interested in collaborations to develop new diagnostic tests.

Bioinformatics
Bioinformatics is significantly simplified with TempO-Seq compared to RNA-Seq. Rather than de novo sequencing every single base, short defined input sequences are measured using sequencing platforms, which increases the efficiency and only needs roughly 10% of the sequencing reads of RNA-Seq for the same sample quality. Alignment time is decreased, and gene counts are delivered to you as .CSV files.
