Sample requirements

TempO-Seq is able to efficiently quantify gene expression for small and (traditionally) “difficult” samples.

The requirements listed below are guidelines rather than rules. The assay has been shown to work with a wide variety of different tissue types, including whole zebrafish larvae, tear drops, and heart biopsies, so even if a tissue is not specifically listen below, it might still be suitable for TempO-Seq analysis. We are happy to discuss supporting your sample types with you - please get in touch and a member of the team will get back to you shortly.

FFPE tissue

Our standard FFPE assay uses between 2 mm squared to 10 mm squared from 4 - 5 µm sections. Typically, we will require an indication of which area of the slide you would like us to analyse (e.g., from an H&E counterstain).

Tissue microarrays

Tissue microarrays are a useful tool to analyse many patient samples at once and are commonly used for immunohistochemistry. We have previously worked with samples taken from tissue microarrays and obtained good data.

LCM samples

Occasionally, higher spatial resolution than 2 mm squared is desired. We have obtained successful results from customer-provided laser-captured microdissected samples.

Cell lysates

The assay runs directly from crude cell lysates, including 3D spheroids. In contrast to many other methods, TempO-Seq has an upper rather than a lower limit of cells for input. Therefore, “low cell numbers” for many other technologies are perfectly fine for use with TempO-Seq. Please get in touch to discuss your project.

Blood

TempO-Seq runs directly from whole blood, buffy coat, and peripheral blood mononuclear cells (PBMCs). We have adapted a simple protocol for collection and storage of whole blood samples for TempO-Seq. Please get in touch to find out more.
